Virussen detecteren en hun = concentratie=20 meten, in niet meer dan enkele minuten. Dat wordt mogelijk met de=20 ultragevoelige sensor die onderzoeker dr. Aurel Ymeti heeft ontwikkeld samen met = dr.=20 Hans Kanger, = prof.dr. Jan=20 Greve en prof.dr. Vinod Subramaniam van de Biophysical Engineering = groep=20 van de Universiteit Twente. Snelle screening in ziekenhuizen of op = luchthavens, om uitbraak van bijvoorbeeld SARS v=C3=B3=C3=B3r te = zijn, wordt met de=20 nieuwe sensor mogelijk; een klein speeksel- of bloedmonster = volstaat. De=20 onderzoekers presenteren hun resultaten in de februari-editie van=20 Nano=20 Letters, en = trekken nu al=20 de aandacht van onder meer MIT=E2=80=99s Technology=20 Review en = Materials Science=20 and Nanotechnology@Nature.com.

De vraag naar een detectiemethode die snel, gevoelig en=20 gemakkelijk in het gebruik is, is actueel in het licht van recente = uitbraken van onder meer SARS en het H5N1 vogelpestvirus. Zeker in = gebieden die niet kunnen beschikken over geavanceerde laboratoria, = is een=20 handheld detector het beste alternatief, maar dit geldt = eigenlijk voor=20 alle plaatsen waar ter plekke een snelle screening vereist is. De=20 onderzoekers zijn erin geslaagd een compact prototype te maken = door twee=20 technieken te combineren: meting van een interferentiepatroon van = licht en=20 antilichaam-antigen herkenning.

Het lichtpatroon dat binnen enkele minuten verschijnt, = geeft=20 meteen informatie over de aanwezigheid en concentratie van = virussen. De=20 detector heeft daarvoor een aantal verschillende lichtgeleidende = kanalen=20 die elk gevoelig zijn gemaakt voor een specifiek virus. Elk kanaal = heeft=20 daarvoor een coating van antilichamen die bij het betreffende = virus horen.=20 Licht, afkomstig uit een laser, wordt uitgesplitst over de = kanalen. Hebben=20 zich virusdeeltjes gehecht aan de antilichamen, dan verandert de = gang van=20 het licht. Deze verandering komt tot uiting in het = interferentiepatroon:=20 een vingerafdruk van de aanwezige virussen en hun concentratie is = het=20 resultaat. Deze methode is extreem gevoelig. Het is zelfs mogelijk = om de=20 binding van =C3=A9=C3=A9n enkel virus deeltje te meten. =

Schematische weergave van = de sensor:=20 monochromatisch licht van een laser bron wordt in een kanaal = golfgeleider=20 gekoppeld en verdeeld over vier parallelle kanalen. Deze vier = kanalen=20 worden gevormd door een referentie kanaal (4) en drie meet kanalen = (1-3)=20 die gebruikt worden om verschillende virussen te meten door op = deze=20 kanalen de juiste antilichamen aan te brengen. Het licht dat uit = de vier=20 kanalen komt interfereert met elkaar om op een CCD camera een=20 interferentie patroon te vormen. Specifieke binding van een virus = aan een=20 van de drie kanalen door middel van de aangebrachte antilichamen=20 veroorzaakt een fase verschuiving van het licht in dat kanaal. = Deze fase=20 verschuiving wordt gemeten als een verschuiving van het = interferentie=20 patroon. Op deze manier geeft een analyse van het interferentie = patroon=20 informatie over de hoeveelheid virus deeltjes die zich op de = verschillende=20 kanalen gehecht hebben.

De onderzoekers stellen in hun publicatie dat de = techniek beter=20 geschikt is dan bijvoorbeeld het bestaande Polymerase Chain = Reaction=20 (PCR): eenvoudiger in het gebruik, sneller, en toch met een = vergelijkbare=20 gevoeligheid. Voordeel is onder meer dat een bloed- of = speekselmonster=20 nauwelijks voorbewerkt hoeft te worden voor de meting. Ook is het = mogelijk=20 om modules voor verschillende virustypen snel uit te wisselen. =

Het prototype waarover nu in Nano Letters wordt = gepubliceerd, is=20 resultaat van het promotie-onderzoek van Aurel = Ymeti. Zijn onderzoek is gefinancierd door de = Technologiestichting STW,=20 en is verricht samen met ondernemingen zoals Paradocs Group BV, = bioM=C3=A9riex=20 BV, en LioniX BV. Het onderzoek van de groep Biophysical = Engineering is=20 ingebed in twee UT-instituten: het MESA+ Instituut voor = Nanotechnologie en=20 het BMTI Instituut voor Biomedische Technologie.
